MicroRNAs (miRNAs) have been reported to regulate the expression of genes in many processes. MiRNA in extracellular vesicles (EV) has also been studied, while no study miRNAs in EVS semen. Subgroup J Avian leukosis virus (ALV-J) can be transmitted vertically, but the mechanism was not clear enough. This study was to examine the expression profile of miRNA in EVs semen and to investigate the relationship between it and the vertical transmission by gene ontology (GO) and pathway enrichment analysis.
Here, we first isolated and characterized EVS semen with Nanoparticle Tracking Analysis, Western Blot and transmission electron microscopy experiments. By sequencing in each EVS miRNA library, a typical 9 differentially expressed miRNA, including six up-regulated and 3 down-regulated, are identified. Target gene prediction, explanation GO and KEGG pathway enrichment analysis shows that the function may be associated with these miRNAs. Overall, these findings will improve our understanding of the content and composition of the miRNA in EVs semen and provide new insights into the critical role of regulation of miRNAs EVS semen in the transmission ALV-J.
