Circulating MicroRNA Expression Profiles in Patients with Stable and Unstable Angina
Objective: High incidence of and deaths from unstable angina (UA) is, for the most part, a consequence of the lack of highly sensitive and specific non-invasive markers. Circulating microRNAs (miRNAs) have been widely recommended as potential biomarkers for various diseases. In this study, we characterized the typical miRNA expression profile in patients with stable angina (SA), UA, and normal coronary arteries (NCA), and identify promising candidates for the diagnosis of UA.
Methods: Serum collected from patients with SA, UA, and NCA who visited the Department of Cardiovascular Diseases Meizhou People’s Hospital. Small RNA sequencing performed on the Illumina HiSeq 2500 platform. MiRNA expression in different groups of patients profiled and then confirmed by that in an independent set of patients. The function of miRNAs are expressed differently allegedly using gene ontology classification and Kyoto Encyclopedia of Genes and Genomes pathway analysis.
Results: Our results suggest that circulating miRNA expression profiles differ between SA, UA, and patient NCA. A total of 36 and 161 miRNAs are dysregulated in SA and UA patients, respectively. validated miRNA
expression by reverse transcription quantitative polymerase chain reaction. Conclusion: The results showed that circulating miRNAs are potential biomarkers UA. There is a positive correlation between birth weight and BMI pregestational pregnant individuals (r = 0.48, P <0.001). One hundred and eight miRNAs were differentially expressed between normal and overweight / obese group, which target genes are enriched in the metabolic pathways.
C19MC five miRNAs (miR-516a-5p, miR-516b-5p, miR-520A-3p, miR-1323, mir-523-5p) were upregulated in overweight / obese group. Target enrichment analysis indicates their involvement in the post-embryonic development, lipid and glucose homeostasis, differentiation of T cells and the development of the nervous system.
Circulating MicroRNA Expression Profiles in Patients with Stable and Unstable Angina
Variant transthyretin amyloidosis (ATTRv) is a rare autosomal dominant disease characterized by the accumulation of amyloid in many organs, most cause sensory-motor neuropathy, cardiomyopathy, and dysautonomia. The purpose of this study was to report the profiles of microRNAs (miRNAs) expression ATTRv identified in patients’ blood.
ATTRv ten patients, 10 asymptomatic carriers of the variant transthyretin (TTRv), 10 patients with Charcot-Marie-Tooth disease (CMT), and 10 healthy controls were studied. Human Schwann cell cultures were used to study the regulation of miR-150-5p effect on the expression of cAMP response element-binding protein (CREB), brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF).
ATTRv patients had 33 miRNAs up-regulated and 48 down-regulated compared to healthy controls; 9 miRNAs up-and-regulated and 30 down-regulated compared CMT patients; 19 miRNAs up-and-regulated and 38 down-regulated compared TTRv asymptomatic carriers. Twelve of the 19 miRNAs upregulated have fold higher than 100. The validation study showed miR-150-5p as a valuable biomarker for patients ATTRv TTRv distinguish from asymptomatic carriers (AUC: 0.9728; p <0.0001).

Schwann cell culture model suggests that miR-150-5p is a strong negative regulator of genes CREB, BDNF and NGF. Identification of deregulated miRNAs may help in understanding the complex underlying pathomechamism ATTRv development and related multisystemic pathology. Further investigation is needed on the role of circulating miR-150-5p to predict shifts TTRv operator of asymptomatic status appearance of symptoms.
